This is the first of two lectures by Phil Green on “Sequence Assembly.” Yeast and some of the bacterial genomes have already been completely sequenced. There is also a rapidly growing body of known human sequence. All this has been because of our ability to read DNA. This lecture discusses the shotgun strategy for rapid DNA sequencing, and begins discussion of sequence assembly algorithms (to be concluded in the next lecture). The method and importance of assigning error probabilities to each base are also discussed.
